Pharmacological action

Broad-spectrum antibiotic. The mechanism of action is associated with the suppression of the first stage of the synthesis of bacterial cell wall peptidoglycan. As a structural analogue fosfoenola pyruvate, enters into a competitive interaction with the enzyme N-acetyl-glucosamine-3-o-transferase enolpiruvil, The result is a specific, selective and irreversible inhibition of this enzyme.

It is active against most Gram-negative bacteria: Pseudomonas aeruginosa, Escherichia coli, Proteus is wonderful, Klebsiella pneumoniae; Gram-positive bacteria: Staphylococcus spp. (incl. Staphylococcus aureus, Staphylococcus epidermidis), Streptococcus spp. (incl. Enterococcus faecalis).

Pharmacokinetics

Absorption sostavlyaet 60%; plasma protein binding – 10%. Cmax when administered in a dose of 50 mg / kg – 32 ug / ml; at / in a dose 20 mg / kg – 130 mg / kg; T1/2 – 2.2 no. Quickly penetrates and well distributed in the tissues. 90% excreted by the kidneys with the creation of high concentrations in the urine.

Testimony

Treatment of infectious and inflammatory diseases (caused by sensitive microorganisms) lower urinary tract, incl. acute bacterial cystitis, exacerbation of chronic recurrent bacterial cystitis, acute bacterial urovezikalny syndrome, bacterial non-specific urethritis, asymptomatic bacteriuria massive (Pregnancy), postoperative urinary tract infection. Preventing urinary tract infections during surgery and transurethral diagnostic surveys.

Dosage regimen

Is the inside. Single dose for adults – 3 g, for children – 2 g. The treatment regimen is established individually, with regard to the stage of inflammation, reactivity of the patient, the sensitivity of microorganisms to fosfomycin.

Side effect

Rarely: nausea, vomiting, diarrhea, skin rash, disappear on their own after the abolition of fosfomycin.

Contraindications

Increased sensitivity to fosfomycin, pregnancy.

Pregnancy and lactation

Fosfomycin is contraindicated during pregnancy.

During lactation the use of fosfomycin is possible only in cases of extreme necessity under the supervision of a physician.

Cautions
